Generating assay-ready proteins depends on two key steps: isolating the protein of interest from complex starting material and ensuring the purified protein is in an appropriate buffer. Here we describe a high-throughput workflow for the purification and buffer exchange of his6-tagged proteins. Using Ni-IMAC IMCStips® followed by SizeX150 IMCStips® on the Analytik Jena CyBio FeliX, his6-tagged GFP was successfully isolated from bacterial lysate with >80% recovery and >95% purity from affinity purification and >96% reduction in small molecule contaminants with SizeX buffer exchange. This streamlined approach provides a reproducible and efficient route to prepare proteins suitable for downstream applications, reducing the variability and labor typically associated with manual workflows.
