Simplifying Urine Drug Testing with “Flash Hydrolysis”

- IMCSzyme® RT hydrolyzed >90% of morphine, oxymorphone, and codeine glucuronides within 15 minutes at room temperature
- Achieved results using just 5 μL, which is 1/20th the enzyme volume of a competing product
- The competing enzyme left significant unhydrolyzed substrate in two of three analytes tested, even at 100 μL (2:1 enzyme-to-urine ratio)
STUDY OVERVIEW
Dr. Tania Sasaki at Altius Diagnostics evaluated hydrolysis efficiency under demanding conditions: 20,000 ng/mL each of morphine, oxymorphone, and codeine glucuronides in 50 µL of urine (60,000 ng/mL of glucuronidated substrate total).
Each enzyme manufacturer's recommendations were used for the hydrolysis conditions: 5 µL for IMCSzyme RT and 100 µL for the competing "all-in-one" premixed enzyme. Remaining glucuronides were monitored at 8-minute intervals at room temperature.
RESULTS
IMCSzyme RT hydrolyzed greater than 90% of all three glucuronides within 15 minutes.
The "all-in-one" premixed enzyme, at 20 times the volume, still had 23% of oxymorphone glucuronide and 60% of codeine glucuronide remaining.
