What is Multi-Attribute Method (MAM) and how would it streamline sample preparation?

Multi-attribute method (MAM) is a mass spectrometry-based technique that enables users to simultaneously monitor and quantify molecular attributes with the potential to impact the efficacy, pharmacokinetics, and safety of biotherapeutic applications. From proof-of-concept and product development to real-time quality control, MAM is intended to consolidate separate assays and streamline laboratory workflows.

Current sample preparation for MAM relies on a manual buffer exchange step to remove excess denaturant and accelerate subsequent trypsin digestion efficiency. Circumventing the buffer exchange step requires significant sample dilution and prolonged digestion time, which may result in sample preparation artifacts. Through the use of SizeX IMCStips on Hamilton Microlab STAR platforms, automated buffer exchange for MAM is an easy and hands-free, walkaway method.

Enhancing the Multi-Attribute Method through an Automated and High-throughput Sample Preparation

Our recent peer-reviewed research published in the journal mAbs describes a completely automated multi-attribute method (MAM) sample preparation approach. For the first time, a complete tip-based MAM sample preparation protocol is automated on a single robotic liquid handling system, leveraging a deck layout that integrates both heating and cooling functionalities. Rapid desalting with SizeX IMCStips® is achieved with Hamilton Robotics. The fully automated method achieves high-throughput sample preparation for MAM, while maintaining superior method performance.

Automated Multi-Attribute Method Sample Preparation using High-Throughput Buffer Exchange Tips

Our latest peer-reviewed research published in the journal Rapid Communications in Mass Spectrometry describes a fully automated multi-attribute method (MAM) sample preparation protocol. Rapid desalting in less than 15 minutes is achieved using miniaturized size exclusion chromatography columns (SizeX IMCStips®) on an automated liquid handler (a Hamilton Microlab STAR). This automated protocol enables the preparation of up to 96 samples with less “at-bench” time than the manual preparation of a smaller batch of samples, thereby greatly facilitating support of process development and the use of MAM in QC.

Gray D. Amick, Ph.D., is the Director of Operations at IMCS with over 26 years of experience in forensic DNA analysis and toxicology. Prior to joining IMCS, he led forensic DNA testing for the Richland County Sheriff’s Department as technical leader and lab director. He has been court-qualified as an expert over 100 times and has authored and co-authored multiple posters and peer-reviewed articles.

Amanda C. McGee is a Research Scientist at IMCS involved with enzyme characterizations, new analytical method developments, and advanced technical support. She joined IMCS with several years of experience in analytical testing for active pharmaceutical ingredients as per cGMP, USP and ICH guidelines. She has co-authored peer reviewed articles in the Journal of Analytical Toxicology and presented research at national and international conferences.
