Reducing Analytical Complexity by the Application of Enzymes

Presenter: Dale Dexter Expand

Dale Dexter is the Managing Director and Principal at DASH Lab Services LLC. His background and expertise originated with working on LCMS systems at a small CRO in the Kansas City area after college, which is also where he met his business partner. Years later, after gaining substantial experience with LCMS systems and the clinical world in general, Dale’s team helps clinical laboratories of all sizes achieve their goals and realize their growth via the offerings of DASH Lab Services. These include method development, validation, report writing, compliance from all aspects, custom Cal and QC solutions, remote data review, technical supervisory and lab directorship fulfillment, and overall clinical lab management and support.

Presenter: John J. Tomashek, Ph.D. Expand

John J. Tomashek earned his Ph.D. in Biochemistry and Molecular Biology at the University of California, Davis, as a member of the Plant Cell Biology Training Group. He has published research in the areas of bioenergetics, multidrug resistance, enzyme structure/function relationships, metagenomics, and science education. For almost ten years he led the enzyme improvement team for a major company developing second generation cellulosic biofuels. He is coinventor of almost two dozen patent families, primarily in the areas of enzyme discovery and engineering. Currently, John works at IMCS to build a platform for developing improved protein and enzyme products.

In Part I, Dale Dexter will go through critical aspects for successful LC-MS/MS separation and quantitation of drug analytes. This will include a review of chromatographic peak deconvolution and identification, the use—and cost—of standards to quantify peak areas, and the overall exponential complexity that occurs as new analytes are added to a panel. In Part II, John Tomashek will introduce enzymes as a way to decrease the complexity and the cost of analysis. He will review the fundamental aspects of how enzymes work and the basics needed for successful enzyme hydrolysis. In Part III, John Tomashek will present the application of different commercial enzymes to real samples to show the sorts of pitfalls that can occur, and you can ensure you are getting consistent valid results.

At the end of this webinar, participants will:

  • Have an increased appreciation for the enormous complexity and challenges of LC-MS/MS separation and quantitation of analytes;
  • Understand how enzymes work and can reduce the complexity of drug analysis, and

Caleb R. Schlachter, Ph.D., as the Principal Scientist at IMCS, leads and provides guidance for several research and development projects that involve proteins, including enzymes for glycan hydrolysis and glycan synthesis. He has co-authored multiple patents, posters, and peer-reviewed articles on β-glucuronidases and sulfatases.

Gray D. Amick, Ph.D.

Director of Operations
Gray D. Amick, Ph.D., is the Director of Operations at IMCS with over 26 years of experience in forensic DNA analysis and toxicology. Prior to joining IMCS, he led forensic DNA testing for the Richland County Sheriff’s Department as technical leader and lab director. He has been court-qualified as an expert over 100 times and has authored and co-authored multiple posters and peer-reviewed articles.

Amanda C. McGee

Research Scientist
Amanda C. McGee is a Research Scientist at IMCS involved with enzyme characterizations, new analytical method developments, and advanced technical support. She joined IMCS with several years of experience in analytical testing for active pharmaceutical ingredients as per cGMP, USP and ICH guidelines. She has co-authored peer reviewed articles in the Journal of Analytical Toxicology and presented research at national and international conferences.
