SizeX IMCStips for Tecan
SizeX IMCStips® are miniaturized size exclusion chromatography columns designed for automated, rapid buffer exchange and desalting. Packed with a hydrophilic polymeric resin, SizeX IMCStips have remarkable chemical and physical stability for various applications. Our tips are designed for seamless compatibility with Tecan Fluent® and Tecan EVO® automation workstations, enabling you to rapidly process eight samples in less than 10 minutes.
SizeX IMCStips effectively isolates green fluorescent protein (GFP) from a mixture of GFP and bromophenol blue (BPB). While BPB is retained in the tips, GFP is eluted after SizeX processing. The absorbance spectra show the difference between the initial mixture containing both GFP and BPB (Pre-SizeX Sample) and the spectra of the purified GFP after processing with SizeX (Post-SizeX). The Starting Sample represents a deconvoluted spectrum of the GFP fraction of the Pre-SizeX Sample. Dotted line indicates that the concentration of small molecule BPB is reduced by more than 256 times.
AUTOMATED BUFFER EXCHANGE WITH SIZEX IMCSTIPS ON A TECAN FLUENT 1080
Protein recovery of various starting concentrations of green fluorescent protein (GFP) ranging from 0.1-1.0 mg/mL (n=2). Automated buffer exchange was performed with SizeX150 and SizeX100 on a Tecan Fluent 1080 with the Flexible Channel Arm (FCA). GFP concentrations were measured by using the absorbance at 505 nm.

Recovery of Green Fluorescent Protein (GFP) from solutions of pure GFP (1 mg/mL) and a combination of GFP with Bromophenol Blue (GFP+BPB; GFP at 1 mg/mL, BPB at 0.1 mg/mL). The samples were processed using SizeX150 IMCStips on a Tecan Fluent 1080 system. Measurements by absorbance at 505 nm and 591 nm and by fluorescence (Exλ=485 nm, Emλ=530 nm) were used to determine yields. The average GFP recovery yield with SizeX150 for GFP+BPB and GFP only samples (n=6) were 94.2±6.1% (absorbance, Table 2) and 96.0±11.4% (fluorescence, Table 3). Yields were calculated using the corrected absorbance at 505 nm after subtracting the average value of BPB (0.7098) and the blank (0.038) from the raw sample.

Protein Recoveries Following Automated Affinity Purification Using IMCStips on a Tecan Fluent
Cell culture media was processed with Affinity IMCStips on a Tecan Fluent. The starting sample volume was 850 μL at a concentration of 337 μg/mL (or 286 μg of starting protein). Measurements were performed using Octet. 150 μL of samples were eluted with 100 mM glycine, pH 3.0, and neutralized with 15 μL of 1 M Tris HCl, pH 8.5.
