Comparative IMCStip-Based Protein A Purification on the Tecan Fluent: Impact of Resin and Elution pH on Antibody Aggregation
Automated optimization of antibody purification is highly advantageous for
enhancing efficiency and reproducibility in protein purification workflows.
In this application note, Affinity and SizeX IMCStips are used on a Tecan Fluent
system to automate the purification and subsequent buffer exchange of
monoclonal antibodies from clarified cell culture supernatants. The evaluation
specifically examines two protein A resins (MabSelect™ PrismA and Praesto™
Jetted A50 HipH) across varying elution pH levels, focusing on recovery and
aggregation impacts. Results demonstrate the influence of resin choice and
pH conditions on purification outcomes, thus highlighting the value of
automating with IMCStips in streamlining process development workflows.
