IMCSzyme® RT is our second-generation enzyme designed to hydrolyze more efficiently in a shorter amount of time and at room temperature. IMCSzyme RT hydrolyzes glucuronidated drugs of abuse in 15 minutes or less at temperatures between 20 to 25 °C. This enzyme is resistant to naturally occurring inhibitors found in human biological samples. This product is a genetically modified β-glucuronidase (CAS: 9001-45-0; EC: 3.2.1.31) that catalyzes the cleavage of β-D-glucuronic acid from drug metabolites. This product contains a superior recombinant form of β-glucuronidase with enhanced activity. Our superior formulation ensures that after opening, this product is stable for up to 12 months.

IMCSzyme RT® (Room Temperature) delivers the fastest hydrolysis available, now in just 15 minutes or less and at room temperature incubation.

This enzyme hydrolyzes a broad substrate of glucuronides efficiently. This superior enzyme is resistant to naturally occurring inhibitors found in human biological samples, which provides your lab with dependable hydrolysis results no matter what.

Enzyme Performance based on Recommended Hydrolysis Reaction

Certified drug-free human urine samples were fortified with 16 glucuronide standards (5,000 ng/mL each when liberated). 50 μL of fortified human urine was mixed with 165 μL of master mix and incubated at room temperature for 15 minutes. The graphs below compare IMCSzyme RT vs. BpGUS and PvGUS enzymes.

  1. IMCSzyme RT hydrolyzes all glucuronidated drugs—including codeine-6-β-glucuronide (C6G)—in less than 15 minutes at room temperature
  2. Compared to other purified enzymes in the market, IMCSzyme RT achieves 100% C6G hydrolysis; others achieve less than 20%
  3. Only IMCSzyme RT consistently outperforms other enzymes across multiple glucuronidated substrates and across different urine types
  4. Presence of natural inhibitors in drug-free urine results in reduced recovery of parent compound for BpGUS and PvGUS compared to Surine™ and synthetic urine standards

Enzyme Performance at pH 4-7

Broad working pH range ensures accuracy even with the most difficult urine samples. IMCSzyme RT comes with the new Room Temperature Buffer (RTB) that is formulated with no phosphate, making this buffer mass spectrometry-friendly.

Gray D. Amick, Ph.D., is the Director of Operations at IMCS with over 26 years of experience in forensic DNA analysis and toxicology. Prior to joining IMCS, he led forensic DNA testing for the Richland County Sheriff’s Department as technical leader and lab director. He has been court-qualified as an expert over 100 times and has authored and co-authored multiple posters and peer-reviewed articles.

Amanda C. McGee is a Research Scientist at IMCS involved with enzyme characterizations, new analytical method developments, and advanced technical support. She joined IMCS with several years of experience in analytical testing for active pharmaceutical ingredients as per cGMP, USP and ICH guidelines. She has co-authored peer reviewed articles in the Journal of Analytical Toxicology and presented research at national and international conferences.
